Raul looked short of match fitness when he played in the second half of Real's 2-1 defeat to Bayern Munich in the final of the club's pre-season centenary tournament on Sunday and coach Vicente del Bosque has left him out of the squad.
Guti is ruled out with an ankle injury suffered in that game.
The game between the Spanish and Italian clubs at the New York Giants stadium in East Rutherford will raise funds for United Nations programmes to fight HIV/AIDS and the Global Fund to Fight AIDS, Tuberculosis and Malaria.
