The 26-year-old, who joined Real from Sao Paulo in December 2005, was carried off on a stretcher in the 29th minute of the league match against Real Betis on Saturday after turning awkwardly while challenging for the ball.
"We think he has torn the anterior cruciate ligament, but we will have to carry out further tests tomorrow," Real Madrid doctor Juan Carlos Hernandez told reporters.
Real host Dynamo Kiev in Champions League Group E on Tuesday having lost their opening match 2-0 away to Olympique Lyon.
