Playas Jandia, based in the Canary Islands, host Real Madrid, embarrassingly knocked out in the second round by Toledo last year, while Figueres have been drawn at home to Barcelona.
"We may not know much about them but they deserve our full respect," Real's Brazilian defender Roberto Carlos said of the lowly opposition.
Figueres are more used to playing against Barcelona's B team in the regional third division but will relish the chance to take on the likes of Rivaldo and Patrick Kluivert.
Cup holders Real Zaragoza will also face third division opposition, having been drawn away to Logrones.
All but two of the one-off ties are scheduled for October 10. Those involving Barcelona and Deportivo Coruna will be played on November 7 to avoid a clash with their re-scheduled Champions League matches.
